FIELD: biochemistry.
SUBSTANCE: invention relates to biochemistry and more particularly to a method for producing an antibody. Method comprises culturing a host cell comprising a polynucleotide comprising a first region of translation initiation (TIR), operably linked to a polynucleotide encoding an antibody heavy chain and second TIR, operably linked to a polynucleotide encoding a light chain antibody and recovering the antibody from the host cell culture. First TIR contains a variant of co-translational prokaryotic secretion signal sequence DsbA and second TIR comprises co-translational or post-translational prokaryotic secretion signal sequence. Due to the presence of said polynucleotide in the host cell heavy and light chains are packed and assembled to form a biologically active antibody. Present invention also discloses a polynucleotide and a host cell to produce antibodies, i.e., to implement the above method.
EFFECT: present invention allows to increase the yield of the resulting antibody.
