FIELD: medicine.
SUBSTANCE: invention refers to medicine, namely transfusiology, and can be used to monitor efficiency of donor blood irradiation. Method involves placement of blood cell blast-transformation reaction with mitogen assay and accounting reaction results by determining lymphocyte stimulation index. Carrying out a reaction is carried out in respect to irradiated donor blood, results accounting is made with help of flow cytofluorometer using monoclonal antibodies, marked different staining, gating of activated lymphocytes with reduced expression of marker general leukocytal CD45 and determining among them percentage of double-negative T-lymphocytes with an immunophenotype CD3+CD4-CD8-, wherein unexposed sample is taken from same blood. Then efficiency of radiation is determined from ratio of content of double-negative T-lymphocytes with immunophenotype CD3+CD4-CD8- in tested irradiated and same unirradiated control blood as Eir=Sdntlo-Sdntlk, where Eir is radiation efficiency Sdntlo is percentage content of double-negative T-lymphocytes with immunophenotype CD3+CD4-CD8- in test (in irradiated tested blood), Sdntlk is percentage content of double-negative T-lymphocytes with immunophenotype CD3+CD4-CD8- in control (unirradiated sample of same blood). If difference with control is more than 50 %, conclusion is made of functional disability of T-lymphocytes in irradiated blood and immunological safety of given blood preparation for recipient.
EFFECT: using given method enables to determine effectiveness of irradiation of donor blood by flow cytometry.
