FIELD: medicinal genetics.

SUBSTANCE: the present innovation deals with the methods for detecting somatic gene mutations in human cells. For implementing the present method it is necessary to isolate the fraction of mononuclear cells of peripheral blood and dye them with labeled antibodies to CD3 and CD4 to analyze cells immunophenotype with the help of flow cytometry. The isolated cells should be additionally stained with labeled antibodies to CD56 and (or) HLA-DR. Then based upon the values of light diffusion one should isolate intact lymphocytes to detect among them mutant CD3-CD4+CD56- (and/or HLA-DR-) cells. The frequency of mutant cells should be calculated as the ratio of the number of CD3-CD4+CD56- (and/or HLA-DR-) lymphocytes against the number of CD3+CD4+ cells. Application of the present innovation enables to increase the accuracy in identifying and quantitative evaluation of mutant cells, carry out dosimetry and indication of the action of genotoxic factors upon human body.
