FIELD: biotechnology.
SUBSTANCE: invention relates to cellular engineering, and can be used to determine the biological activity of IFN-γ. A human melanoma cell line PiGAS with the registration number VKPM H-161 is obtained by transfection of MelP parent human melanoma cell line with pGAS(x6)-Luc//Neo plasmid encoding 6xGAS-Luc reporter construct consisting of sequence of SEQ ID NO: 1, comprising 6 repetitions of the STAT-1 transcription factor binding site, minimal SV-40 promoter, and Photinus pyralis (Luc) gene, as well as sequence SEQ ID NO: 2, comprising the Neo resistance gene to the selective antibiotic G418 under the control of its constitutive PGK-1 promoter.
EFFECT: obtaining of a reporter cell line that allows determination of bioactive IFN-γ by of STAT-1 - signaling pathway activation detection.
