Abstract RU 2192467 C2

FIELD: biochemistry, genetic engineering, enzymes. SUBSTANCE: invention relates to preparing proteins with luciferase activity and enhanced thermal stability as compared with natural enzyme. Method of preparing involves substitution of glutamic acid residue in conservative region 354-361 in amino acid sequence of luciferase from Photinus pyralis or in corresponding region of enzyme showing more than 60% homology of sequence with luciferase from Photinus pyralis with any other amino acid residue except for glycine, proline and aspartic acid. Modified forms of luciferases comprising additionally hydrophobic amino acid residue in position corresponding to 215 amino acid residue in sequence of luciferase from Photinus pyralis are preferable. Invention proposes DNA sequences providing the preparing indicated modified forms of luciferases by their expression in host-cell that can be used at temperatures above 30 C. Invention can be used in luminometric analysis for measurement of ATP levels. EFFECT: improved method of preparing, valuable properties of enzyme. 15 cl, 15 dwg, 1 tbl
