FIELD: biotechnology.

SUBSTANCE: isolated antibody or antigen-binding fragment thereof is described that binds to the chemokine 4 receptor (CXCR4) and contains: a) heavy chain variable region (VH) containing (i) VH CDR1 selected from the group consisting of SEQ ID NO: 107, 113, 114; (ii) VH CDR2 selected from the group consisting of SEQ ID NO: 162, 128, 154, 123, 158, 124, 159, 125, 160, 126, 161, 127, 163, 164, 165, 166, 167, 168, 155, 129, 156 and 130, and (iii) VH CDR3 selected from the group consisting of SEQ ID NO: 112; and; b) light chain variable region (VL) containing (i) a VL CDR1 selected from the group consisting of SEQ ID NO: 144, 138, 141, 142, 143, 146, 147, 148, 149 and 150; (ii) VL CDR2 selected from the group consisting of SEQ ID NO: 145, 132; and (iii) VL CDR3 selected from the group consisting of SEQ ID NO: 139, 140 and 170. Invention also relates to antibody-drug conjugates comprising such an antibody, nucleic acids encoding the antibody, and methods for producing such an antibody. Invention also relates to therapeutic methods for using said antibody and anti-CXCR4 antibody-drug conjugates for the treatment of a disorder associated with the function or expression of CXCR4 (for example, cancer), such as colon cancer, RCC, esophageal cancer, stomach cancer, head and neck cancer, lung cancer, ovarian cancer, pancreatic cancer, or hematological cancer.
