FIELD: biotechnology.
SUBSTANCE: invention relates to biotechnology, analytical chemistry and method of determining content β-casein A1 and/or A2 groups in milk, dairy products, milk composite products, milk-containing products. Method comprises concentration β-casein in analyzed sample until content is achieved β-casein in a sample of not less than 80 wt. % relative to content of other proteins, fermentative hydrolysis of the obtained concentrate, chromatographic analysis of the obtained hydrolyzate and chromatogram analysis. Enzymatic hydrolysis of step (a) β-casein is carried out using as the enzymes TRSK-treated trypsin in an amount providing a hydrolysis reaction. Chromatographic analysis of the obtained hydrolyzate reveals peaks on a chromatogram corresponding to specific peptides characterizing total content β-casein in hydrolyzate and content β-casein A1 and/or A2 groups, based on which groups in the sample are determined. Specific peptides are determined based on chromatograms obtained for hydrolysates of samples β-casein A1 group and/or β-casein A2 group, wherein chromatographic analysis is carried out at wavelength of 280 nm.
EFFECT: possibility of determining content of A1 and/or A2 β-casein in the sample over a period of time not exceeding 4 hours while simplifying the determination procedure.
