SUBSTANCE: invention relates to biochemistry, in particular to a method for modifying a target genomic locus on a Y-chromosome in a murine embryonic stem cell (ES). Method involves: providing a mouse ES cell containing a target genomic locus on a Y-chromosome, wherein the target genomic locus comprises a recognition site for the nuclease agent, and where the murine ES cell is found in the culture containing the DMEM base medium, the introduction into the mouse ES cell of the nuclease agent or the polynucleotide encoding the nuclease agent, wherein the nuclease agent induces a one- or double-strand break on the recognition site, and a large targeting vector comprising a polynucleotide insert, and identifying at least one mouse ES cell containing a polynucleotide insert integrated into the target genomic locus in its genome.

EFFECT: invention enables efficient modification of the target genomic locus on a Y-chromosome in a mouse embryonic stem cell.
