FIELD: biotechnology.

SUBSTANCE: invention relates to screening of a target compound for feminizing activity in XY pluripotent cells of a non-human mammal, optimizing concentration of the target compound in a feminizing environment and creating a donor XY pluripotent cell of a non-human mammal. Method comprises culturing a first population and a second population of XY pluripotent cells of a non-human mammal in a medium containing a basic medium and an additive, suitable for maintaining pluripotency of XY pluripotent cells of a non-human mammal, wherein the first population is cultured in the presence of the desired compound, and the second population is cultured in the absence of the desired compound. That is followed by analyzing one or more XY pluripotent cells of a non-human mammal in each of the first and second populations of XY pluripotent cells of a non-human mammal with respect to expression and/or activity of one or more of Ddx3y, Uty and Eif2s3y, according to which feminizing activity is determined by reducing expression and/or activity of one or more of Ddx3y, Uty and Eif2s3y in one or more XY pluripotent cells of a non-human mammal from a first population compared to one or more XY pluripotent cells of a non-human mammal from a second population.
