FIELD: biochemistry.

SUBSTANCE: invention relates to the field of biochemistry, in particular, to a method for targeted modification of a genomic locus of interest in one or more pluripotent rat cells, comprising introducing into pluripotent rat cells a large targeting vector (LTVEC) comprising an insert nucleic acid, and identifying a genetically modified pluripotent rat cell comprising the targeted genetic modification in the genomic locus of interest, and also to a method of creating a humanised rat using said modification method. Also disclosed is a method for targeted modification of a genomic locus of interest in a population of pluripotent rat cells to obtain a genetically modified rat, including providing a population of pluripotent rat cells, obtaining a clone of a pluripotent rat cell, introducing said clone of pluripotent rat cells into a rat host embryo, and incubating said rat host embryo.

EFFECT: invention makes it possible to efficiently obtain a genetically modified rat.
