FIELD: chemistry.

SUBSTANCE: invention relates to a method of producing vesicles with a transmembrane pH gradient. Said method is characterized by the following steps: a) obtaining vesicles consisting of at least one matrix substance in an aqueous medium having osmolarity of not more than 200 mOsm/l, where the matrix substance is selected from a group consisting of amphiphilic lipids forming liposomes, b) mixing the vesicles with a basic or acidic buffer having osmolality of at least 200 mOsm/l higher than the osmotic activity of the aqueous medium of step a), for applying osmotic shock to the vesicles and producing the vesicles filled with a buffer, and c) diluting a mixture of an aqueous medium and a basic or acidic buffer containing vesicles filled with a buffer by adding a neutralizing solution to obtain vesicles with a transmembrane pH gradient, suspended in a suspension buffer, where the suspension buffer differs from the basic or acidic buffer from the pH value.

EFFECT: method according to the invention provides stability of the vesicles obtained after step a) during sterilization by autoclaving.
