FIELD: biotechnology.
SUBSTANCE: invention relates to the field of biotechnology. Invention is a test system for identifying tissue DNA of rats (Rattus) and mice (Mus musculus) in dry fodder and meat semi-products, including a buffer for polymerase chain reaction, mixture for its carrying out, consisting of deoxynucleoside triphosphates, oligonucleotide primers and fluorescent probes, specific for section of animal's genome and for internal control sample; mixture of enzymes from DNA polymerase with antibodies inhibiting enzyme activity, TAQ POLYMERASE, an internal control sample in the form of a bacteriophage T4 suspension with concentration of 5×103 phagic particles per 1 mcl, a negative control sample which is a mixture of recombinant plasmid DNA, containing a fragment of the animal's genome and a fragment of the bacteriophage T4 genome with a nucleotide sequence: T4F TACATATAAATCACGCAAAGC – forward primer, T4R TAGTATGGCTAATCTTATTGG – reverse primer, T4P FAM ACATTGGCACTGACCGAGTTC BHQ1 – probe, according to the invention, phagolysate of bacteriophage T4 is used for internal control sample, and for positive control sample fragments of genomes of native bacteriophage T4 and rats (Rattus) and mice (Mus musculus) taken in volume ratio 1:1:1 with the following nucleotide sequence are used: Rat-F: 5'-GCCTTCCTACCATTCCTGCAT-3' – forward primer, Rat-R: 5'-AGGAGGTTGGCTACTAGGAT-3' – reverse primer, Rat-P: 5'-FAM-ACGCAGCTTAACATTCCGCCCA-3'-BHQ1 – probe, Mus-F: 5'-GTTGCTTTGTCTACTGAG-3' – forward primer, Mus-R: 5'-ACCTGAAACATTGGAGTA-3' – reverse primer, Mus-P: PAM-5'-TCGCAGTCATAGCCACAGCA-3-BHQ1 – probe.
EFFECT: invention allows to expand functional capabilities, increase accuracy of identification of species identity.
