FIELD: biotechnology.
SUBSTANCE: invention relates to biotechnology. The invention is aimed at identifying rat (Rattus) and mouse (Mus musculus) tissues spices in dry feed and meat semi-finished products, including the isolation of DNA from animal tissues by sorption, setting up a polymerase chain reaction with fluorescent detection with 45 real-time amplification cycles using site-specific animal DNA genomes, oligonucleotide primers, probes, fluorescent dyes: JOE/Yellow for a specific signal for one of the animals and Cy5/Red - for internal control sample of a suspension of bacteriophage T4 with a concentration of 5×103 phage particles per 1 µl of positive control sample in the form of a mixture, containing fragments of the genomes of animals and bacteriophage T4 nucleotide sequence, taken in the ratio 1:1:1, and measuring the accumulation of fluorescent signals on channels of appropriate fluorescent dyes, the interpretation of the results on the basis of the presence or absence of crossing of fluorescence curve with the threshold line, if the curves of the accumulation of the fluorescent signal out to 35 cycle, the reaction is considered positive, and if the curves do not cross the threshold line or cross it after 35 cycles, then the result of the reaction - negative, according to the invention, DNA isolated from tissues of rats (Rattus) and mice (Mus musculus) and internal control sample using the phage lysate of phage T4, and for the positive control sample - fragments of the genomes of native bacteriophage T4 and tissues of rats (Rattus) and mice (Mus musculus), for the determination of DNA in tissues of the rat (Rattus) using the fluorescent dye JOE/Yellow, and for the fabric of the mouse (Mus musculus) - FAM/Green.
EFFECT: said invention allows increasing accuracy of species identification, simplifying the preparation process.
