SUBSTANCE: described is a group of inventions involving a method of producing a heteromultimer protein comprising a first polypeptide comprising a hinge region having a first heterodimerization domain, where the first polypeptide containing the hinge portion is connected to the first light chain, and a second polypeptide comprising a hinge portion having a second heterodimerization domain, where the second polypeptide containing the hinge portion is connected to the second light chain, where the second heterodimerization domain interacts with the first heterodimerization domain on the interaction surface, and where the first and second polypeptides containing the hinge region are connected by at least one interchain disulfide bond (versions). In one embodiment, the method comprises culturing host cells capable of expressing a first and a second polypeptide, and obtaining a combined culture medium for a first host cell and a second host cell without destroying a cell membrane of host cells, wherein the combined culture medium comprises a heteromultimer protein.
EFFECT: invention extends range of methods for producing heteromultimer proteins.
