FIELD: biotechnology.
SUBSTANCE: group of inventions relates to designing a biocatalyst strain having nitrilase activity and high operational stability. Disclosed is a method of constructing on the basis of Rhodococcus bacteria a biocatalyst strain having nitrilase activity and high operational stability, by integrating into the genome of the recipient strain of bacteria of the genus Rhodococcus two copies of the sequence SEQ ID 01, which is a nitC1 nitrilase gene from the Alcaligenes denitrificans strain C-32 VKM B-2243D. Also disclosed is a recombinant strain of Rhodococcus rhodochrous bacteria VKPM AS-2111, which is a biocatalyst having nitrilase activity. Said strain is obtained by integrating two copies of the nitC1 nitrilase gene from the Alcaligenes denitrificans strain C-32 VKM B-2243D into the recipient strain Rhodococcus rhodochrous VKM Ac-1515D. Said biocatalyst strain is used in the method of synthesis of acrylic acid in form of ammonium acrylate.
EFFECT: invention discloses a method for constructing a biocatalyst strain based on bacteria of the genus Rhodococcus, having nitrilase activity and high operational stability, recombinant strain of Rhodococcus rhodochrous bacteria obtained by this method, a method for synthesis of acrylic acid using this strain as a biocatalyst.
