FIELD: biotechnology.

SUBSTANCE: method for assessing the reproductive activity of influenza viruses in a trivalent and quadrivalent live influenza vaccine, based on a one-step real-time RT-PCR using hydrolysable oligonucleotide fluorescent probes. Real-time RT-PCR using the developed specific primers and oligonucleotide probes is sensitive and specific for studying reproduction of vaccine viruses in MDCK cell culture in a trivalent or quadrivalent vaccine. Use of real-time RT-PCR makes it possible to study mutual influence of components of polyvalent LIV even at reduced level of reproduction of vaccine viruses in cells of mammals in comparison with reproduction in CE. Developed method enables assessing reproduction of influenza A and B vaccine viruses in a trivalent vaccine in samples obtained from respiratory tracts of laboratory animals and patients after immunization with trivalent LIV.

EFFECT: method enables assessing the mutual influence of influenza B viruses in Victoria and Yamagata antigenic lines for inclusion in polyvalent vaccines.
