METHOD FOR DIAGNOSING INFLAMMATORY PROCESS IN HUMAN MYOCARDIUM Russian patent published in 2021 - IPC G01N33/58

Abstract RU 2750905 C1

FIELD: medicine, cardiology in particular.

SUBSTANCE: invention relates to the field of medicine, in particular to cardiology and laboratory diagnostics, and is intended for the diagnosis of the inflammatory process in the human myocardium. Antibodies to human myocardial tissue antigens are determined by conducting an indirect immunofluorescence study of bovine myocardial tissue. Bovine myocardial tissue taken from the left ventricle from the endocardium is examined. Pre-freezing is carried out by placing the fabric in a container with isopentane cooled to a temperature of (-145)-(-159)°C, while the temperature of the isopentane is kept constant, the freezing time is 10-40 s. Fabric sections with a thickness of 4 microns are prepared in a cryostat, after which the sections are dried at room temperature and fixed with acetone at +4°C. The patient's blood serum is titrated to 1:160-1: 320 and incubated on sections for 1.5-2 hours at a temperature of (+36)-(+37) °C. If the study reveals the presence of a reaction in at least one of the zones, namely, in the wide anisotropic disks of cardiomyocytes, in the endothelial zone of capillaries or fibers of the conducting system of the heart, the presence of an inflammatory process is diagnosed.

EFFECT: invention provides an increase in the accuracy of detecting the inflammatory process in the human myocardium and a reduction in the time of the study.
