SUBSTANCE: group of inventions is described, including closed-end linear duplex DNA (hereinafter – ceDNA) for the delivery of heterologous nucleic acid, a composition for the delivery of heterologous ceDNA to a subject, a host cell for the delivery of heterologous ceDNA to a subject, a method for the delivery of heterologous ceDNA to a mammalian cell, a method for the delivery of heterologous ceDNA to a subject, where the method involves the delivery of ceDNA to a subject, and the delivery of ceDNA does not lead to an immune response against ceDNA in the subject, a method for the delivery of ceDNA to a subject, where the method includes the delivery of the host cell and the method for obtaining ceDNA. In one implementation, ceDNA contains an insert of heterologous nucleic acid, where the specified insert is flanked by at least two sequences of inverted terminal repeats (ITR) of the adeno-associated virus (AAV).
EFFECT: obtaining closed-end linear duplex DNA for non-viral gene transfer.
