SUBSTANCE: inventions relate to the field of biotechnology, specifically to gene-therapeutic drugs based on proteins with antitumor and immune-modulating effects; they can be used for the treatment of human oncological diseases. A genetic cassette is proposed, containing codon-optimized nucleotide sequences of TRAIL, PTEN and IFNβ-1 genes represented with SEQ ID NO:1, SEQ ID NO:2, and SEQ ID NO:3, respectively, separated with a nucleotide sequence of a row of P2A, or E2A, or F2A, or T2A peptide, for expressing genes encoding TRAIL, PTEN and IFNβ-1 proteins with the possibility of being used in a lentiviral eukaryotic expression vector. A pharmaceutical composition is also proposed for the treatment of a human oncological disease containing mesenchymal stem cells obtained from patients with an oncological disease expressing a genetic cassette TRAIL-P2A-PTEN-P2A-IFNβ-1, TRAIL-E2A-PTEN-E2A-IFNβ-1, TRAIL-F2A-PTEN-F2A-IFNβ-1 or TRAIL-T2A-PTEN-T2A-IFNβ-1 in the effective amount.
