FIELD: biotechnology.
SUBSTANCE group of inventions relates to biotechnology and immunology, in particular, to hinge structures in antigen-binding constructs. Proposed is an amino acid hinge region containing EPKSSDKTHTCPPCPPC. Also proposed is an amino acid hinge region containing the central hinge sequence CPPCPPC (SEQ ID NO: 52) or CPPCPPCPPC (SEQ ID NO: 54) attached to the upper hinge sequence ELKTPLGDTTHT (SEQ ID NO: 48) or EPKSSDKTHT (SEQ ID NO: 46). The above hinge regions are located in mini-antibodies binding to the target molecule. The mini-antibodies contained in the composition can be applied for diagnostic visualisation or delivery of a therapeutic payload.
EFFECT: inventions provide an improved construct of a mini-antibody containing the described hinge region, provide a possibility of improving the stability and/or reducing the aggregation of mini-antibodies, reducing the level of half-molecules to less than 1%.
