FIELD: biotechnology.
SUBSTANCE: group of inventions relates to biotechnology and immuno-oncology, in particular, to a bispecific binding protein binding to PD-1 and CTLA-4. The protein comprises a first heavy chain including CDR1 - GFTFSDYGMH, CDR2 - YISSGSYTIYSADSVKG, CDR3 - RAPNSFYEYYFDY; a first light chain including CDR1 - SASSKHTNLYWSRHMYWY, CDR2 - LTSNRAT, CDR3 - QQWSSNP; a second heavy chain including CDR1 - GFTFSSYGMH, CDR2 - VIWYDGSNKYYADSVKG, CDR3 - DPRGATLYYYYYGMDV; and a second light chain including CDR1 - RASQSINSYLD, CDR2 - AASSLQS, CDR3 - QQYYSTP. Also proposed are a composition containing such a protein, a nucleic acid molecule encoding the protein, a host cell for expression thereof, a method for treating or preventing cancer in a subject, and a method for increasing the immune response in a subject with cancer or subjected to a risk of developing cancer.
EFFECT: invention allows for production of bispecific binding proteins with improved binding properties relative to the initial monospecific binding proteins targeting the same molecules, even if used in combination.
