FIELD: biotechnology.
SUBSTANCE: present invention relates to a method for producing particles of a recombinant adeno-associated viral vector (rAAV) with a high level of selection or with a high title. The method includes: collection of cells and/or cell culture supernatant, containing particles of rAAV vector; their optional concentration; lysis; filtration; exposure of the resulting lightened lysate to ion-exchange chromatography on a column, and optionally concentration of the resulting eluate; mixing of eluate from the column or concentrated eluate from the column with cesium chloride, and exposure of the resulting mixture to gradient ultracentrifugation to separate particles of a full-fledged rAAV vector from particles of empty AAV capsid and other impurities related to AAV vector; collection of particles of the full-fledged rAAV vector separated at the previous stage, and obtaining a composition of particles of the full-fledged rAAV vector in a buffer with non-ionic surfactant; exposure of particles of the full-fledged rAAV vector obtained at the previous stage to buffer exchange by means of flow filtration along the flow with subsequent filtration.
EFFECT: implementation of the invention provides for the improvement of extraction and/or increase in the title of particles of the full-fledged rAAV vector after the purification process.
