METHOD FOR PREPARATION OF MATURE NEMATODES FOR IDENTIFICATION BY MALDI TOF MASS SPECTROMETRY Russian patent published in 2022 - IPC G01N33/483

Abstract RU 2768162 C1

FIELD: medicine.

SUBSTANCE: invention refers to medicine, namely to parasitology. Fragment of a large nematode (roundworm, dirofilaria) or a whole small nematode (pinworm), pre-washed once in solution of 0.9 % NaCl, is homogenized in a clean disposable plastic container with a sterile pestle. Further, 50 mcl of the obtained mass is moved by the dispenser into an Eppendorf test tube and is partially dehydrated by centrifugation for 11 minutes in the RPM = 13300 rpm mode (RCF = 9888 g), after removal of the supernatant liquid fraction by the dispenser, 30 mcl of the standard OS solution is added. Obtained suspension is mixed on vortex for 1 minute, 30 mcl of 70 % formic acid is added to it, the suspension is repeatedly mixed on vortex for 1 minute and centrifuged for 11 minutes at RPM = 13300 rpm (RCF = 9888 g). After removing the liquid supernatant with a pipette, the obtained precipitate is deposited using a disposable loop on a smooth target, dried at room temperature, coated with a CHCA (α-Cyano-4-hydroxycirmamicacid), remains at room temperature until dry; then identification on MALDI ToF mass-spectrometer follows.

EFFECT: simplified and accelerated sample preparation.
