FIELD: pharmaceuticals.
SUBSTANCE: proposed group of solutions relates to a ginkgo biloba leaf extract providing an anti-PAF (platelet activation factor) effect and nerve nutrition. Alcoholic extract of ginkgo biloba leaves, providing an anti-PAF (platelet activation factor) effect and nerve nutrition, produced from ginkgo biloba leaves by alcohol extraction and complying with the following conditions: 1) the content of rutin is less than or equal to 4.0 wt.%; 2) the content of quercetin is less than or equal to 0.4 wt.%; 3) the content of bilobalide is from 2.6 to 4.8 wt.%; 4) the content of ginkgolide J is from 0.1 to 0.5 wt.%; 5) the content of residual ethanol is less than or equal to 0.5 wt.%; 6) the content of biflavonoids is less than or equal to 0.02 wt.%, the biflavonoids include amentoflavone, bilobetin, and ginkgetin; 7) the content of genistin is 0, and the content of ginkgolide M is 0; wherein alcohol extraction includes the following stages: A) crushing the dried leaves of ginkgo biloba seedlings to produce the crushed ginkgo biloba leaves; C) adding a 60% aqueous solution of ethanol to the crushed ginkgo biloba leaves, performing double extraction by heating under reflux, each time for 3 hours, and producing a first filtrate by filtration; C) adding water to the filtered residue of the medicinal product remaining after filtration, performing a single extraction by heating under reflux with an extraction time of 0.5 hours, and then producing a second filtrate by filtration; D) combining and concentrating the second filtrate and the first filtrate into a thick paste, dissolved in purified water heated to 60-100°C, leaving to cool and filtering, resulting in a filtrate; E) concentrating the filtrate to a relative density of 1.04 to 1.08 and absence of an alcohol odour, adding purified water in an amount 1.5 to 2.5 times greater than the amount of powdered ginkgo biloba leaves, subjecting to inter-layer cooling in cooling water at 5 to 7°C for 12 to 24 hours, and executing centrifugation of the supernatant liquid at a speed of 13,000 to 15,000 rpm to produce a centrifugal liquid; F) loading the centrifugal liquid into a column with macroporous resin followed by direct flushing with purified water for 1 to 2 hours and back flushing for 0.5 to 1.5 hours, consecutively eluting with 18% aqueous ethanol solution, a 30% aqueous ethanol solution, and a 50% aqueous ethanol solution resulting in 18% ethanol eluent, a 30% ethanol eluent, and a 50% ethanol eluent, respectively, constituting first eluents; in the first eluents, the 50% ethanol eluent constitutes a first eluent with a high solvent concentration, and the combined solution of the 18% ethanol eluent and the 30% ethanol eluent constitutes a first eluent with a low solvent concentration; G) concentrating the first eluent with a low solvent concentration to eliminate the alcohol odour, loading the first eluent with a low solvent concentration into a polyamide column, subjecting to direct flushing with purified water in a volume 0.5 to 2 times greater than the volume of the polyamide column, followed by elution with ethanol resulting in an ethanol eluate constituting a second eluate; H) combining and concentrating the second eluent and the first eluent with a high solvent concentration until the alcohol odour is eliminated; I) extracting the concentrate with cyclohexane, discarding the cyclohexane extract, spray drying the concentrated solution or concentrating the concentrated solution into a thick paste and vacuum drying the thick paste; grinding the dried products, passing through a sieve of 80 to 100 mesh and mixing in a common mixer at a speed of 10 to 12 rpm for 1.5 to 2.5 hours, resulting in an alcoholic extract of ginkgo biloba leaves. Method for producing an alcoholic extract of ginkgo biloba leaves. Application of the production method for producing an alcoholic extract of ginkgo biloba leaves. Medicinal agent based on the alcoholic extract of ginkgo biloba leaves, providing an anti-PAF effect and nerve nutrition, produced from the alcoholic extract of ginkgo biloba leaves.
EFFECT: above extract is characterised by improved quality: high content of active components and low content of harmful components.
