SUBSTANCE: invention relates to the field of oil well research and can be used in the development of oil fields. According to the method, when drilling exploration, pilot or production wells, the composition of traces of microorganisms characteristic of various layers of rock through which the borehole passes at this field is determined by sampling the sludge during drilling and then determining the composition of traces of living or dead microorganisms using DNA analysis, while from the samples of drilling sludge and/or drilling mud, by direct seeding and cumulative cultivation, strains of microorganisms inherent in drilling sludge/drilling mud are isolated and determined, to which type of medium: water-saturated or oil/ gas-saturated, the detected microorganisms belong, and by the composition of traces of microorganisms detected in the borehole fluid, the source or layer of the incoming borehole fluid is determined.

EFFECT: simplifying the method for monitoring the origin of the extracted borehole fluid.
