FIELD: biotechnology; diagnostical medicine.
SUBSTANCE: invention relates to the field of biotechnology and diagnostical medicine, and is a new method for semi-quantitative analysis of certain molecules of microRNA (hereinafter – miRNA) in samples of RNA isolated from biological (biopsy) material. The method includes sequential conductance of two reactions: reverse transcription (hereinafter – RT) and polymerase chain reaction (hereinafter – PCR), wherein a sequence of a reverse PCR primer provides its interaction only with a section of complementary DNA, synthesized as a result of RT reaction, eliminates a possibility of interaction with RT primer and initiation of PCR after inefficient RT in the absence of analyte (miRNA).
EFFECT: invention allows for significant increase in analytical sensitivity of a technology of analysis of miRNA, using two-tailed RT-PCR, which defines a possibility of use of this technology for medical diagnostics.
