FIELD: biotechnology.
SUBSTANCE: substance of the proposed invention consists in using the phage Yersinia enterocolitica 2021 (FC-115) as a diagnostic agent, wherein the phage is applied in the form of a track on a two-layer lawn, the lower layer whereof constitutes a 1.5% Hottinger agar medium in Petri dishes and the upper layer is produced from the strain under study, pre-grown on a Hottinger agar (pH 7.2) in Petri dishes; the culture is then plated in 4.5 ml Hottinger broth and placed in a thermostat for 24 hours at 28 °C until a final concentration of n·10 8 m.c./ml is reached, followed by adding 0.5 ml of the broth culture to 4.5 ml 0.7% Hottinger agar, premelted and cooled to 45 °C; then a second layer of the resulting mixture is poured onto the surface of the lower layer, and after the upper layer solidifies, the bacteriophage Y. enterocolitica 2021 (FC-115) is applied; the plated cultures are incubated in a thermostat for 24 hours at 28 °C, followed by visually registering the result based on the presence of a lysis zone on the culture under study and making a conclusion on the presence of the Y. enterocolitica pathogen.
EFFECT: invention can be used in laboratory diagnosis of yersiniosis caused by Yersinia enterocolitica for the identification and differentiation thereof.
