METHOD FOR IDENTIFICATION OF CHOLERA VIBRIO O1 SEROGROUPS OF BIOVARS CLASSICAL AND EL TOR Russian patent published in 2020 - IPC C12Q1/02 C12Q1/70 C12R1/63

Abstract RU 2729575 C1

FIELD: medical microbiology.

SUBSTANCE: invention refers to medical microbiology, particularly to laboratory diagnostics, and can be used for development of new diagnostic preparations allowing to identify cholera strains O1 of a serogroup of biovars Classical and El Tor. Method provides using the microbial lysability with a diagnostic phage on an agar medium. Bacteriophage strain Rostov-6 is applied, which is applied in the form of a path on the surface of a petri dish filled with a prepared solution consisting of the analyzed culture, which is introduced into Martin's broth and left in thermostat for 4 hours at temperature of 37 °C. Then the specified amount of mixture is taken and added to 0.7 % of melted and cooled to 45 °C of Marten agar. Mixture is mixed in a test tube in circular motions and quickly poured onto the surface of a petri dish with 1.5 % agar. It is evenly distributed over the surface and allowed to dry. After drying, the preparation is coated with a drop of the bacteriophage Rostov-6 preparation, the inoculation is incubated in a thermostat at temperature of 37 °C for 24 hours. That is followed by identification on the grown culture by the presence of a phagolysis zone, which confirms the presence of cholera vibrio O1 serogroups of biovars Classical and EL Tor or their absence.

EFFECT: invention allows to increase the degree of authenticity of identification of cholera vibrio of 01 serogroup Classical and El Tor.
