FIELD: biotechnology; molecular genetic diagnostics; genetics and breeding of farm animals.
SUBSTANCE: invention helps to assess allelic polymorphism of the beta-casein (CSN2) gene of cattle by a molecular genetic method of research. In the method of genotyping cattle for the A1 and A2 alleles of the CSN2 beta-casein gene by real-time PCR, the intercalating dye SYBR green is used as a fluorophore, which, by the presence of successful amplification in the presence of an S-shaped amplification curve and with a difference in the threshold Ct cycle in sample down from the negative control by more than 1.5 cyclesΔ Ct > 1.5 makes it possible to unambiguously determine the presence of each of the studied alleles of the CSN2 gene in the analysed DNA sample and, accordingly, the genotype of the animal.
EFFECT: simplification of the method of genotyping cattle for the A1 and A2 alleles of beta-casein, reduction of the time of analysis and significant reduction of the cost of analysis by eliminating the use of allele-specific fluorescently labelled probes and replacing them with the intercalating universal dye SYBR-GREEN.
