SUBSTANCE: invention relates to the field of biotechnology, molecular genetic diagnostics, genetics and selective breeding of farm animals, in particular, to assessment of the allelic polymorphism of the kappa casein gene (CSN3) of cattle by the method for molecular genetic research. Proposed is a method for PCR with allele-specific probes for genotyping cattle by alleles a and b of the kappa-casein gene. The following are used in the method: a direct common primer CSN3-RT-F-114 with the nucleotide sequence 5′-agagcccacctgagatcaac-3′; a reverse common primer CSN3-RT-R-114 with the nucleotide sequence 5′-tcttggctgttattcattttgc-3′; a B-allele-specific fluorescence-labelled probe Probe 1 with the nucleotide sequence FAM-caactgcggtctaaatactctaaggag-BHQ1; an A-allele-specific fluorescence-labelled probe Probe 2 with the nucleotide sequence HEX-caactgcagtctaaaaactctaaggag-BHQ1.

EFFECT: expanded the range of methods for genotyping cattle by alleles A and B of the kappa-casein gene (CSN3) based on real-time PCR and higher reliability thereof.
