METHOD OF DETERMINATION OF GLUTATHIONE IN WHOLE BLOOD Russian patent published in 2023 - IPC G01N27/26 G01N1/28

Abstract RU 2799875 C1

FIELD: medicine.

SUBSTANCE: method of determining glutathione in whole blood is disclosed including venous blood sampling, hemolysis, protein precipitation, capillary electrophoresis with spectrophotometric detection of peaks at a wavelength of 200 nm with processing of the analysis results, the results of which are used to judge the concentration of reduced and oxidized forms of glutathione, while during hemolysis, water, acetonitrile with N-ethylmaleimide at a concentration of 10–30 mmol/l and ethanol at a ratio of 2:1:3:3 are added to the venous blood sample, respectively, mixed, centrifuged, and then glycylasparagine is added in water at a concentration of 1 mmol/l and diethyl ether in the amount of 0.2 and 1.08 volumes of the supernatant, respectively, centrifugation is carried out, then water is added to the separated sedimentary residue in a volume of 75% of the volume of the venous blood sample, while during capillary electrophoresis, first into a capillary with an internal diameter 50 microns with a total length of 39.5–42.5 cm, citric acid solution at a concentration of 75 mmol/l, 5% of the volume of the capillary, the prepared sample is sequentially injected, and then sodium tricitrate is injected at a concentration of 75 mmol/l, while electrophoresis is carried out with a voltage of 12–14 kV, 75 mmol/l of sodium citrate with a pH of 5.8, containing 200 μmol/l of cetyltrimethylammonium bromide and 5 μmol/l of sodium dodecyl sulfate, is used as a background electrolyte.

EFFECT: invention provides an increase in the accuracy, reliability and reproducibility of the analysis results.
