SUBSTANCE: invention is related to polynucleotide component 5'UTR of a eukaryotic expression vector, and can be used for in vitro RNA synthesis, increasing its translational efficiency in eukaryotic cells and regulating gene expression. The polynucleotide component 5'UTR of the eukaryotic expression vector contains the leader regulatory sequence TPL (three-part leader sequence) with SEQ ID NO:5.

EFFECT: invention provides increased expression of a sequence of interest in eukaryotic expression systems through the use of said TPL regulatory leader sequence.
