SUBSTANCE: present invention relates to cellular technologies, and can be used to express a polypeptide of interest in CHO cells. By combining a specific polynucleotide sequence 5'UTR and the secretory leader sequence hCD33 in an expression cassette for the expression of a polypeptide of interest and further transformation of the obtained host cell genetic construct, a CHO cell is produced with a higher expression level of the polypeptide of interest as compared to the prototype expression cassettes.

EFFECT: invention makes it possible to produce a polypeptide of interest in high yield.
