FIELD: medicine; immunology.

SUBSTANCE: invention concerns a method of determining specific sensitization to antigens of uveoretinal tissue, cornea and lens. The method includes a cell migration screening test. Water-salt extracts of uveoretinal tissue, lens, and cornea of bovine eyes, initially containing 3.0–5.0 mg/ml of total protein, are used as antigenic preparations. Antigenic medicinal products are added to the culture medium in the following working concentrations: 50 μl/ml of uveoretinal tissue extract, 45 μl/ml of cornea extract, 45 μl/ml of lens extract, and to 100 μl of the culture medium containing the test antigen in the working concentration or control without antigen, 5 µl of leukocyte suspension with a concentration of 2–4×106 cells/ml isolated from the patient’s peripheral blood are added. Then incubation is carried out at 37°C for 18–20 hours. The results of the reaction are taken into account by determining the migration areas based on the average diameter of the circle formed by the microculture for microcultures in the experiment (Sop.) and control (Scontr.), and the migration index (MI) of leukocytes is calculated using the formula MI = Sop./Scontr. When the MI value is less than 0.8 calculated units or the MI value is more than 1.2 calculated units, the presence of specific sensitization to antigens of uveoretinal tissue, cornea and lens is determined.

EFFECT: invention provides a simple, accessible, economical method of diagnosing organ-specific autoimmunization in eye diseases, easily reproducible in clinical diagnostic laboratories.
