FIELD: biotechnology.

SUBSTANCE: following is described: a nucleic acid encoding a mutant of the capsid protein of an adeno-associated virus (AAV), which includes a peptide containing an amino acid sequence selected from the group consisting of the following SEQ ID NO: 18, 15–17 and 19–62. Recombinant DNA for producing an AAV particle having tropism for the brain, containing the specified nucleic acid is also described. A cell for producing an AAV particle having tropism for the brain, containing the specified nucleic acid or recombinant DNA is presented. Also the following is provided: AAV particle for gene insertion into the brain containing a mutant of the AAV capsid protein that includes a peptide containing an amino acid sequence selected from the group consisting of the following SEQ ID NO: 18, 15–17 and 19–62. The following is disclosed: a method of producing a cell transduced with a gene, where the method includes the step of contacting an AAV particle containing a mutant of the AAV capsid protein, which includes a peptide containing an amino acid sequence selected from the group consisting of the following SEQ ID NO: 18, 15-17 and 19-62, with a cell.

EFFECT: method of producing an AAV particle is provided, wherein the method includes isolating or purifying the AAV particle from a culture supernatant or cell lysate.
