SUBSTANCE: present application relates to gene therapy and molecular biology. Invention discloses an isolated modified protein VP1 of the capsid of adeno-associated virus serotype 5 (AAV5), which contains one or more amino acid substitutions compared to the VP1 protein of the wild-type AAV5 capsid, which increase the efficiency of transduction, higher efficiency of packaging viral genomes of AAV with preparations based on rAAV5, as well as higher efficiency of production (assembly) of vector based on recombinant adeno-associated virus serotype 5 (rAAV5). Said amino acid substitutions are selected from: a) G226V, b) S2A, G226V and T711S, c) T614A, d) S2A, T614A and T711S, e) T614V, or f) S2A, T614V and T711S, where the amino acid sequence of the wild-type AAV5 capsid VP1 protein has the amino acid sequence presented by SEQ ID NO: 1.

EFFECT: invention can be used in genetic engineering when producing vectors for delivering genetic products to a subject in need thereof.
