FIELD: chemistry.

SUBSTANCE: invention relates to a method of producing unfractionated heparin. Proposed method envisages use of frozen pig intestinal mucosa as raw material and involves raw material defrosting; its homogenisation and extraction with 1.2 M aqueous solution of sodium chloride at temperature of 80±5 °C for 1.0±0.1 hours. Then, cooling is carried out to temperature of 20±2 °C; separation of the extract by centrifugation; filtering the extract and diluting it with water. Aqueous solution is passed through a column with a macroporous anionite pre-balanced with water; anionite is washed with 0.60 M aqueous solution of sodium chloride at pH 2.5±0.1, then 0.65 M aqueous solution of sodium chloride at pH 2.5±0.1. Heparin is eluted with 1.6 M aqueous solution of sodium chloride at pH 2.5±0.1; the eluate is concentrated and purified in an ultrafiltration apparatus in a tangential flow with a molecular weight exclusion limit of 5,000 Da. After that, anhydrous sodium acetate is added to the concentrate to a concentration of more than 7 wt.% and precipitated with methanol to a final concentration of methanol of 37±1% at temperature +4±1 °C for 18±2 h. Precipitate is washed with methanol and the precipitate is separated by centrifugation. Then sodium heparin is dried under vacuum at temperature of 60±3 °C.

EFFECT: invention increases output of heparin, its specific activity, reduces time and simplifies the method of producing.
