FIELD: chemistry.
SUBSTANCE: invention relates to biochemistry, in particular to a method of producing antigen-binding proteins against a human antigen of interest, which involves obtaining a genetically modified mouse in which expression of the autoantigen is eliminated. Production of said mouse involves introduction of Cas9 protein or nucleic acid coding Cas9 protein into a mouse or rat embryo, as well as a first and a second guide nucleotide sequence.
EFFECT: invention makes it possible to effectively reduce the time required for obtaining a genetically modified mouse with low tolerance to a foreign antigen of interest, and also enables to obtain a large number of antibodies against a foreign antigen of interest.
