SUBSTANCE: disclosed are compounds having the structure (I):

or a pharmaceutically acceptable salt or isomer thereof, wherein R1 is phenyl or 5-9 membered heterocyclyl containing 1-3 heteroatoms selected from N and O, wherein R1 is optionally substituted with one or more Rq; each Rq is independently C1-6 alkyl, phenyl, C3 cycloalkyl, 5–6 membered heterocyclyl containing 1–3 heteroatoms selected from N and S, -OR, -C(O)OR, -C(O)R, -X, -CX3, -CX2H, -C(X)H2, -CN, -N(R)2, -N(R)C(O)R, -N(R)S(O)2R, -S(O)2R or -(CH2)nQ’, where Q' is X, -CX3, -CX2H, -CN; each R is independently H, C1-6 alkyl, phenyl or 5-6 membered heterocyclyl containing 1-2 heteroatoms selected from N, O and S; R2, R3, R4, R5 and R6 independently represent H, C1-6 alkyl, X, -CX3, -CX2H or -CN; each Rx independently represents H; W is N; Z is or CRz; Rz is H; each X is independently F, Cl, Br or I; and each n is independently equal to 1 or 2, as well as a pharmaceutical composition based thereon and a method of treating a MRGPR X2 dependent or MRGPR X2 ortholog dependent condition.
