METHOD FOR PRODUCTION OF ACID PHOSPHATASE Russian patent published in 1995 - IPC

Abstract SU 1529725 A1

FIELD: microbiological industry. SUBSTANCE: strain Asp. foetidus F-164 is used to prepare desired product. Culture medium which is used for its cultivation contains (mass): maize starch 4-10, wheat bran 0.05-0.125, aminosubtiline having activity 600 units per 1 g 0.02-0.05, ammonium sulfate 0.4-1.0, magnesium sulfate 0.025-0.0625, potassium chloride 0.025-0.0625, manganese chloride 0.005-0.0125. Seeding of microorganism is carried out at 40-45 C, cultivation is carried out at 35-50 C, rate of aeration being 1-1.5 volumes of air per 1 volume of culture liquid per 1 minute. Stage of active biosynthesis of ferment takes place at 28-30 C at rate of aeration 1 volume of air per 1 volume of culture liquid per 1 min. Thus prepared culture liquid is then filtered, the process is followed by sterile filtration, ultraconcentration and spray drying. 1 of magnesium sulfate as stabilizing agent is added into ultraconcentrate before its drying is carried out. At the final stage of fermentation autolysis of culture takes place. Thus activity of acid phosphatase in culture liquid increases by 20-25 waste amount being decreased 3 times. Activity thus prepared acid phosphatase is up to 180000 units per 1 g, its yield being 10 kg/m3. EFFECT: increases activity and yield of desired product. 3 tbl
