FIELD: medicine, particularly, experimental allergology. SUBSTANCE: group of controls were preliminarily three times on alternate days subjected to base sensibilization using horse serum as foreign protein which was administered intra-abdominally each time in dose of 0.5 ml. In case of other group of animal controls, base sensibilization was accompanied by intra-abdominal administration three times each alternate day of xenobiotic solvent using syringe each time holding dose of 0.5 ml. Compounds to be tested for sensibilizing properties were intra-abdominally administered three times on alternate days into experimental group of animals against background of sensibilization triggered by foreign protein of horse serum taken in test dose and admixed with 0.5 ml solvent. After elapse of fourteen days from the date experiment was started, mast cells of peritoneal exudate of animals were isolated. Glass slide was wetted with 0.3-percent solution of neutral red dissolved in absolute alcohol and then 0.05 ml of peritoneal suspension and 0.05 ml of horse serum were applied. Droplets of both substances were gently mixed and spread over glass surface. The whole was incubated in thermostat during 15 min at +37 C and then air-dried. Reaction results were assessed by calculating one hundred mast cells and determining percentage of cells that reacted to allergen. Test results were considered as negative if number of degranulating cells in experiment did not exceed 10 % as compared with controls. EFFECT: disclosed method allows reproducing natural metabolism of xenobiotics in organism, excluding possibility of direct cytotoxic effect owing to the fact matter under test has no direct contact with mast cells and testing fat- and alcohol-soluble compounds.
