FIELD: medicine. SUBSTANCE: polypeptide is isolated from venum of spider Agelenopsis aperta by chromatography at the range 20-230 nm on column type VidacR C-18 22x250 mm, pore size is 300 , particle size is 10 mcm. Column is eluted with a mixture of acetonitrile and 0.1% aqueous trifluoroacetic acid at gradient concentration of acetonitrile by the following programm: 0-30 min - 5-20% acetonitrile, 30-35 min - 20-70% acetonitrile in 0.1% trifluoroacetic acid. Rate flow is 15 ml/min. At 41.5 min fraction is collected which is subjected for chromatography on the same type column 10x250 mm with particle size 5 mcm. Column is eluted at the flow rate 3.5 ml/min with isocratic system of solvents consisting of 70% 0.1% aqueous trifluoroacetic acid and 30% acetonitrile at the same wavelength range. Fraction is collected at 7.5 min which is subjected for lyophilization. Polypeptide with mol. mass 5201 is obtained, and its amino-terminal amino acid sequence is determined. EFFECT: improved method of polypeptide preparing.
