FIELD: biotechnology, genetic engineering, biochemistry. SUBSTANCE: Bacillus strain - a producer of hydrolytic enzyme (in part, Bac. novo sp. PB 92 a producer of highly alkaline protease or Bac. licheniformis T5 - a producer of α-amylase). Genome of this strain has the inserted DNA sequence that incodes the end product. Producer is treated with recombinant plasmid involving the second copy of the indicated DNA sequence, marker gene and temperature-sensitive region of replication onset, or genome DNA of the strain recipient with the sequence inserted in chromosome encoding the end enzyme. The obtained clones were grown on medium with antibiotics and forms containing two copies of DNA sequence encoding hydrolytic enzyme and separated with sequence essential for the host cell were taken.
