FIELD: bio- technology, biochemistry, molecular biology, antibiotics. SUBSTANCE: invention relates to method for preparing vector based on technology of recombinant DNA. Vector is able to be inserted into chromosomal DNA of the strain Penicillium chrysogenum and provides expression DNA sequences from Cephalosporium acremonium encoding enzymes expandase/hydrolase and acetyltransferase under control of promoter IPNS Penicillium. As result of transformation with indicated vector of the strain Penicillium chrysogenum able to produce isopenicillin N recombinant strain is obtained where biosynthesis of all enzymes catalyzing the conversion of adipate to adipoyl-7-amino-cephalosporanic acid occurs. The example of such strain is Penicillium chrysogenum/pTS-8 (ATCC 74187). Adipoyl-7-aminocephalosporanic acid preparing by culturing the recombinant strain in the presence of adipate is treated then with adipoylamidase to form 7- aminocephalo-sporanic acid. Invention provides the possibility to prepare 7-amino-cephalosporanic acid in industrial scale by effective and economy method.
