METHOD OF PREPARING MULTIPARTIAL VACCINE FOR LEPTOSPIROSIS CONTROL IN ANIMALS Russian patent published in 1997 - IPC

FIELD: biotechnology, microbiology, vaccines. SUBSTANCE: method involves selection of leptospirae of the following serogroups: Icterhemorrhagia, Tarasovi, Pomoni or Grippotyphosis, Tarasovi, Pomoni, Seiro or Icterhemorrhagia and Canicola. Leptospirae were cultured on nutrient media based on serum blood sterilized by microfiltration on diaphragms (pore size is 0.2-0.28 mcm). The grown cultures were inactivated with 0.1-0.3% formalin solution, mixed at the definite ratio and concentrated by ultrafiltration on hollow fibers by 20-30 times. Aluminium hydroxide gel is added to concentrate and then protective drying medium containing sucrose, peptone and phosphate buffer is added at the definite ratios. The obtained mixture is subjected for additional inactivation with merthiolate followed by lyophilization. Vaccine exhibits good solubility, harmless, sterility, high activity, convenient at transport and storage. Vaccine is used for leptospirosis prophylaxis.
