FIELD: immunoassays. SUBSTANCE: T- lymphocytes are isolated from venous blood of patient and cultivated for 48 h on a nutritive medium with phytogemagglutinine added. Then colchicine is added to its final concentration 0.5 mcg/ml and lymphocytes are cultivated for further 2 h. Culture broth is centrifuged, supernatant is treated by hypotonic solution and centrifuged once more. Centrifuged lymphocytes are fixed and their preparations are prepared on microscope slides. Thereafter, one calculates the number of lymphocytes containing at least five acrocenters in one acrocenter association of chromosomes and the number of lymphocytes containing no indicated association. One calculates then percentage of the two groups of lymphocytes to total number of lymphocytes being in metaphase state. When percentage of the lymphocytes containing no acrocenter association is increased by 20% compared to the same indication found for the given patient earlier and in presence of lymphocytes containing at least five acrocenters, one estimate activity of T-lymphocytes as satisfactory. When percentage of the former is increased at least by 60% and the latter are absent or when percentage of the former is reduced by at least 60% and content of the latter is increased more than twice, activity of T-lymphocytes is estimated as understated.
