METHOD OF PREPARING CHLAMYDIUM LUMINESCENT DIAGNOSTIC PREPARATION, CHLAMYDIUM LUMINESCENT DIAGNOSTIC PREPARATION AND METHOD OF DIAGNOSIS OF CHLAMYDIUM INFECTIONS Russian patent published in 1999 - IPC

Abstract RU 2141666 C1

FIELD: medicine, immunology. SUBSTANCE: invention proposes a method of preparing the chlamydium luminescent diagnostic preparation based on by-step immunization of mice with an antigenic material as a suspension consisting of a mixture of live chlamydium cells of different species. After immunization ascite sarcoma cells are administrated to mice. An immune ascitic fluid is collected and gamma-globulin fraction is isolated from its followed by its labeling and ampoulizing. Obtained conjugate is dried. The suspension consisting of the mixture of all developing forms of live chlamydium cells of the species Chl. psittaci, Chl. trachomatis, Chl. pneumoniae is used. At the final stage of immunization a double dose of an antigenic material is administrated in animals by intraperitoneal route. Invention proposes also the chlamydium lyophilically dried luminescent diagnostic preparation based on murine gamma-globulins to chlamydium cells conjugated with fluorochrome. As murine gamma-globulins to chlamydium cells the preparation has murine gamma-globulins to chlamydium cells of species Chl. psittaci, Chl. trachomatis, Chl. pneumoniae of their forms of development and additionally it can has the contrasting agent. Also, invention proposes a method of diagnosis of chlamydium infections by treatment of analyzed material with the claimed diagnostic preparation and the following assay of the presence of chlamydium antigens of the different development forms. An acute stage of chlamydium infection or exacerbation of chronic chlamydium infection is diagnosed in the case of the presence of elementary bodies only. The persistent chlamydium infection is diagnosed simultaneously in the case of the presence of elementary bodies, reticular bodies and cytoplasmatic inclusions. The chronic latent chlamydium infection is diagnosed in the presence persistent form only. Invention can be used in clinical and laboratory practice. EFFECT: enhanced effectiveness of diagnosis of chlamydium infections. 7 cl, 1 tbl, 3 ex
