METHOD FOR EVALUATING LYMPH TOXICITY OF DRUGS IN SPONTANEOUS E-ROSETTE-FORMING REACTION Russian patent published in 2000 - IPC

FIELD: medicine. SUBSTANCE: method involves using lymphocyte suspension separated from patient blood treated with heparin, by means of centrifuging on 1.077 density gradient. Incubation of lymphocyte suspension separated from blood lasts 90 min with drug tested for toxic properties to be added. Then, after 2 min long centrifuging, incubation is continued with sheep erythrocytes added during 18 h. Index of lymphocyte sensitivity to the tested preparation is calculated as lymph toxicity index from a formula: LTI= (% quantity of E-rosette-forming complex in experiment) / (% quantity of E-rosette-forming complex in control sample). The LTI <1.0 being the case, conclusion is drawn concerning inhibited lymphocyte E-receptors motility. The LTI value being greater than or equal to 1.0, conclusion of no lymph toxicity available is to be drawn. EFFECT: high accuracy and sensitivity of the method; enhanced effectiveness in differentiating toxic and auto immune states.
