METHOD FOR REVEALING PLAGUE MICROBE IN FLEAS Russian patent published in 2000 - IPC

Abstract RU 2160781 C1

FIELD: medical microbiology. SUBSTANCE: invention is intended for bacteriologic diagnostics of objects suspicious of being plague infection transmission entities. For that, flea suspension in 0.9% sodium chloride solution is prepared and divided into three parts. One part (0.8-1.2 ml) is examined in polymerase chain reaction (0.3-0.4 ml) to detect gene pFra, pCad, pPst sequences. Second part of suspension is sowed (each time 0.1-0.15 ml) on dense Jackson-Berrose nutrient medium with gentiane violet (0.02-0.03 mcg/ml) to detect feature PGm+ (determined by chromosome gene sequence). Third part of flea suspension is sowed (each time 0.1-0.15 ml) on Chottinger agar with gentiane violet (0.02-0.03 mcg/ml) to determine sensitivity to antibiotics using paper disk technique. EFFECT: accelerated plague microbe revealing procedure and enabled determination of virulence of microbes. 3 ex
