METHOD OF PREPARING HIGHLY PURIFIED AGAR AND AGAROSE FROM RED ALGA AHNFELTIA TOBUCHINSKAYA Russian patent published in 2002 - IPC

FIELD: biotechnology. SUBSTANCE: invention relates to processing of agar-carrying algae and production of gel-forming substances from red algae used in scientific researches in biochemistry, immunochemistry, molecular biology, microbiology and for production of streptomycin, penicillin and preparation included to the check list of important medicinal agents in Russian Federation. Method of processing of red alga ahnfeltia tobuchinskaya involves preliminary treatment of alga in calcium hydroxide in concentration 1-2% as measured for calcium oxide at heating up to 100-105 C and hydromodulus = 1:(12-15) for 60-90 min followed by increase of the reaction mixture temperature to 110 C and keeping at this temperature for 60-90 min. Then extraction is carried out 3-fold being the first extraction is carried out at temperature 115-120 C for 6-6.5 h at hydromodulus 1:(10-12) without addition of calcium oxide and highly purified agar is obtained. The second extraction is carried out at temperature 115-120 C for 4-4.5 h at hydromodulus 1:(6-6.5) with addition of calcium oxide taken in the amount 4.0-4.5% to air-dry alga. The third extraction is carried out at temperature 115-120 C for 2.0-2.5 h at hydromodulus 1:(3-3.5) with addition of calcium oxide taken in the amount 2.0-2.5% to air-dry alga mass. Extracts of the second and third extraction are mixed, filtered and subjected for gelatinization. The prepared gel is cut, washed out by infusion in fresh water and then in distilled water for 10-12 h at hydromodulus 1:(2-3). The purification of agar and agarose extracts is carried out in Nutsch filter in a filter partition formed by a layer of filtering diatomite powder with a layer thickness 0.5-1.0 cm and filtering cardboard at temperature 80-85 C. Method ensures to decrease the total process duration of extraction up to 13 h and decrease the extraction fold to 3. The obtained extracts are not mixed: extract after the first extraction is fed to the production of highly purified agar; the second and the third extracts are fed to the production of agarose.
